Description
The mammalian Na+/H+ exchanger isoform one (NHE1), encoded by Solute Carrier Family 9, member 1 (SLC9A1), consists of 12 membrane domains and a cytosolic C-terminal domain. The human protein is 815 amino acids in length. Five hundred N-terminal amino acids make up the transport domain of the protein. SLC9A1 encodes an isoform of the amiloride-sensitive Na+-H+ exchanger that is present in many cells and the basolateral membrane of renal epithelia. NHE1 plays an important role in maintaining intracellular pH homeostasis by exchanging one intracellular proton for one extracellular sodium ion. Expression of NHE1 is modulated in response to chronic metabolic acidosis, growth factors, and phorbol esters. Mice with a homozygous null mutation in Slc9a1 (Nhe1) exhibited ataxia, recurrent seizures, and selective neuronal cell death
