SET and MYND Domain 1 (SMYD1) is a cardiac-and muscle-specific histone methyltransferase that methylates histone H3 at lysine 4 and regulates gene transcription in early heart development. SMYD1 is critical for both embryonic and adult heart development and function in both mice and men. The unique domain structure characterized by a "split" SET domain, a conserved MYND zinc finger, and a novel C-terminal domain (CTD) distinguishes SmyD1 from other SET domain-containing methyltransferases. SMYD1 is a key regulator of myogenic differentiation and acts as a downstream target of muscle regulatory factors, serum response factor (SRF), and myogenin. Zebrafish SmyD1 is a SET and MYND domain-containing protein that plays an important role in myofiber maturation and muscle contraction.
