Description

GRM1 gene, which is located on 6q24.3, encodes the metabotropic glutamate receptor type 1 (mGluR1), a transmembrane protein highly expressed in cerebellar Purkinje cells. Pathogenic variants in GRM1 have been reported only three times in humans, causing autosomal-recessive cerebellar ataxia with an early-onset and intellectual disability or dominant forms of cerebellar ataxia with a less severe phenotype in adults. GRM1 cause a recessive form of cerebellar ataxia, SCAR13. A neuronal receptor, GRM1, when ectopically expressed in melanocytes, induces in vitro melanocytic transformation and spontaneous malignant melanoma development in vivo in a transgenic mouse model. GRM1 signaling has been implicated in benign and malignant disorders including prostate cancer (PCa). GRM1 encodes the metabotropic glutamate receptor 1 (mGluR1), whose documented role as a modulator of neuronal signaling and synaptic plasticity makes it a plausible schizophrenia candidate
