WNT1 mutations in humans are associated with a new form of osteogenesis imperfecta and with early-onset osteoporosis, suggesting a key role of WNT1 in bone mass regulation. Mutations in WNT1 compromise activity of the osteoblasts leading to disturbed bone mass accrual, fragility fractures, and progressive skeletal abnormalities. Wnt1, as a regulator of bone formation and remodeling, provides the basis for the development of Wnt1-targeting drugs for the treatment of osteoporosis. Biallelic loss-of-function mutations in WNT1 cause severe osteogenesis imperfecta (OI) with conspicuous involvement of the spine. OI is a heritable skeletal disorder characterized by bone fragility and low bone mass. Heterozygous missense variants in WNT1 are responsible for early-onset osteoporosis with variable bone phenotypes
