Cyclin G-associated kinase (GAK), also known as auxilin 2, is a member of the Numb-associated kinase (NAK) family. Other members include adaptor-associated kinase 1 (AAK1), BMP2-inducible kinase (BIKE/BMP2), and myristoylated and palmitoylated serine/threonine kinase 1 (MPSK1). GAK is ubiquitously expressed in various tissues and is primarily known for its role in clathrin uncoating. At its N-terminus, GAK has a highly conserved serine/threonine kinase domain followed by a lipid-binding PTEN-like domain, a clathrin-binding domain, and a J-domain, the latter two domains being sufficient for Hsc70 and GAK-mediated uncoating of clathrin-coated vesicles. As a serine/threonine kinase, GAK exerts part of its functional effects by targeting downstream substrates.
