Nuclear receptor corepressor 1 (NCOR1) is a transcriptional regulator bridging repressive chromatin-modifying enzymes with transcription factors. NCOR1 is one of those co-repressors that regulate the transcription by facilitating the recruitment of HDAC1, 2, 3, 4, 5, and 7. NCOR1 mediates epigenetic silencing of gene expression in a wide range of developmental and homeostatic processes in many tissues, including the brain. NCOR1 regulates the balance between histone acetylation and histone deacetylation. NCOR1 is listed as one of the 3, 230 dose-sensitive genes which very rarely show truncating mutations in the pediatric population without severe diseases, even in a heterozygous state. NCOR1 mediates ligand-independent actions of the thyroid hormone receptor (TR) during development and in hypothyroidism. NCOR1 is a critical coregulator of the TH receptor (TR) isoforms
