MAP4K3 (also named germinal-center kinase-like kinase, GLK) is a serine/threonine kinase, which belongs to the mammalian Ste20-like kinase family. MAP4K3 is required for robust cell growth in response to amino acids. MAP4K3 positively regulates preterm skeletal muscle satellite cell myogenesis, but may not regulate mTORC1 activity. MAP4K3 may play a role in mTORC1 full activation in response to leucine. MAP4K3 is required for phosphorylation of known mTOR targets such as S6K1 (S6 kinase 1), and overexpression of MAP4K3 promotes the rapamycin-sensitive phosphorylation of these same targets.MAP4K3 is a new component in the nutrient-responsive pathway for mTOR activation and reveals a completely new function for MAP4K3 in promoting cell growth
