Description
Phosphatidylglycerophosphate (PG-P) synthase catalyzes the synthesis of PG-P from CDP-diacylglycerol and sn-glycerol 3-phosphate and functions as the committed and rate-limiting step in the biosynthesis of cardiolipin (CL). The PEL1 gene (now renamed PGS1) encodes the enzyme that catalyzes the committed step of cardiolipin (CL) synthesis. PGS1 forms oligomers and associates with CLS1 and PTPMT1. Cardiolipin and CLS1 are not required for PGS1 to assemble in the complex. PGS1, has no catalytic activity on its own, but is implicated in the localisation of the enzyme at major sites of polyglutamylation, i.e. neurones, axonemes and centrioles. Disruption of PGS1 results in loss of the mitochondrial anionic phospholipids phosphatidylglycerol (PG) and CL
