Description
The histone chaperone FACT (facilitates chromatin transcription) is a large heterodimeric complex with roles in transcription, replication, and repair. FACT promotes and subsequently restricts access to DNA as a result of dynamic nucleosome reorganization. The human FACT complex is composed of two polypeptides identified as Spt16 (Suppressor of ty 16) and SSRP1 (structure-specific recognition protein 1) that are both essential for nucleosome reorganization. The Spt16 protein is divided into three distinct structural domains and a highly acidic intrinsically disordered C-terminal domain required for alleviation of nucleosome blockage during polymerase II elongation.
