Peroxisome proliferator-activated receptor gamma coactivator 1alpha (PGC1alpha) encoded by the PPARGC1A gene is a vital regulator of glucose and fatty acid oxidation, mitochondrial biogenesis, and skeletal muscle fiber conversion. PGC-1alpha is a transcriptional co-activator that has been implicated in the pathogenesis of neurodegenerative disorders. As a central regulator of mitochondrial biogenesis and metabolism, PPARGC1A acts as a powerful coactivator of many transcriptional factors that relate to steroidogenesis. The mitochondrial regulator PPARGC1A regulates mitochondrial biogenesis, reducing senescence of vascular smooth muscle cells (VSMCs). PPARGC1A upregulates autophagy reducing senescence by an SQSTM1-dependent mechanism.
