With a molecular mass of about 1 MDa, mitochondrial complex I is the largest multi-subunit complex of the respiratory chain. Complex I couples electron transfer from NADH to ubiquinone with transmembrane proton pumping contributing to the proton motive force used for ATP synthesis. Complex I contributes significantly to the formation of mitochondrial reactive oxygen species (ROS) and is implicated in the pathogenesis of numerous hereditary and degenerative disorders. Mitochondrial complex I from a number of species exhibits a regulatory switch between a deactive D-form and an active A-form.
