Western blot was performed using Anti-nNOS Recombinant Rabbit Monoclonal Antibody (Product # 703331) and a ~160 kDa band corresponding to nNOS was observed across relevant tissues models tested. Tissue lysates (30 µg lysate) of Mouse skeletal muscle (Lane 1), Mouse cerebellum (Lane 2), Rat skeletal muscle (Lane 3) and Rat cerebellum (Lane 4) were electrophoresed using Novex® NuPAGE® 3-8% Tris-acetate gel (Product # EA0375PK2). Resolved proteins were then transferred onto a nitrocellulose membrane by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:500 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 1:5000 dilution). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
