For immunofluorescence analysis, induced pluripotent stem (iPS) cells were fixed and permeabilized for detection of endogenous FGF4 using Anti-FGF4 Recombinant Rabbit Monoclonal Antibody (Product # 703064, 1:100) and labeled with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A27034, 1:2000). Nuclei (blue) were stained using ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). Panel a-c) clearly demonstrates cytoplasmic localization of FGF4 in iPS cells. Panel (d-f) shows no specific staining in Embryoid bodies which has reduced expression of FGF4, demonstrating specificity. The images were captured at 60X magnification.
