For immunofluorescence analysis, PC-12 cells were fixed and permeabilized for detection of endogenous TYRO3 using Anti-TYRO3 Recombinant Rabbit Monoclonal Antibody (Product # 703295, 1:100 dilution) and labeled with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A27034, 1:2000). Nuclei (blue) were stained using ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). Panel a-c) shows representative undifferentiated cells that were stained for detection and localization of TYRO3 protein (green) with less signal compared to panel d-f) clearly demonstrating enhanced membrane localization of TYRO3 in PC-12, differentiated to neuron-like cells with NGF (200 ng/mL, 7 days). The images were captured at 60X magnification.
