Western blot was performed using Anti-SOD2 Recombinant Rabbit Monoclonal Antibody (12) (Product # MA5-29578) and ~20 kDa band corresponding to SOD2 was induced after TNFα treatment in A549 cells and Hydrogen peroxide treatment in HeLa cells and was also observed in Hep G2. Whole cell extracts (40 µg lysate) of A549 (Lane 1), A549 treated with TNFα (20 ng/mL for 72 hrs) (Lane 2), HeLa (Lane 3) HeLa treated with Hydrogen peroxide (50 µM for 6 hrs) (Lane 4) and Hep G2 (Lane 5) were electrophoresed using NuPAGE® 4-12% Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001).The blot was probed with the primary antibody (1:1,000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4,000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
