Western blot was performed using Anti-RNASE3 Polyclonal Antibody (Product # PA5-79927) and an 18 kDa band corresponding to RNASE3 was observed across Mouse Thymus, Mouse Spleen and Mouse Bone Marrow and was not observed in Mouse Brain and Mouse Testis which are reported to be negative. Tissue extracts (30 µg lysate) of Mouse Thymus (Lane 1), Mouse Spleen (Lane 2), Mouse Bone Marrow (Lane 3), Mouse Brain (Lane 4) and Mouse Testis (Lane 5) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (0.5 µg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4,000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005). Uncharacterized bands were observed at ~30 kDa and ~110 kDa for certain tissue models.
