Western blot was performed using Anti-CD49c (Integrin alpha 3) Polyclonal Antibody (Product # PA5-79528) and a 120 kDa band corresponding to CD49c (Integrin alpha 3) was observed in U-87 MG, A549, A-431 and MDA-MB-231 which are reported to be positive and not in other cell lines like Daudi, HEK-293 and K-562. Membrane enriched extracts (30 µg lysate) of U-87 MG (Lane 1), A549 (Lane 2), A-431 (Lane 3), MDA-MB-231 (Lane 4), Daudi (Lane 5), HEK-293 (Lane 6) and K-562 (Lane 7) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (0.2 µg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036,1:4,000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
