Western blot analysis of MAL was performed by loading 50 µg of mouse mpkCCD and Xenopus 2f3 whole cell lysates per well onto an SDS-PAGE gel. Proteins were transferred to a nitrocellulose membrane and blocked with 5% nonfat dry milk in TBS for 1 hour at room temperature. The membrane was probed with a MAL polyclonal antibody (Product # PA1-085) at a dilution of 1:500 overnight at 4C, followed by an HRP-conjugated goat anti-rabbit IgG secondary antibody for 1 hour.
