Western blot was performed using Anti-PCBD1 Polyclonal Antibody (Product # PA5-66416) and a 12 kDa band corresponding to Pterin-4-alpha-carbinolamine dehydratase was observed across cell lines and tissue extracts tested and not in MOLT-4, Reh, Mouse Spleen and Mouse Brain which is reported to be negative. An uncharacterised band (*) at ~`45 kDa was also observed across cell lines tested. Whole cell extracts (30 µg lysate) of Caco-2 (Lane 1), Hep G2 (Lane 2), A549 (Lane 3), U-2 OS (Lane 4), MOLT-4 (Lane 5), Reh (Lane 6) and tissue extracts of Mouse Spleen (Lane 7), Mouse Brain (Lane 8), Mouse Kidney (Lane 9), Mouse Liver (Lane 10) and Rat Liver (Lane 11) were electrophoresed using Novex™ 16% Tricine Protein Gel (Product # EC66952BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (0.2 µg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076).
