Western blot was performed using Anti-SRC2 Polyclonal Antibody (Product # PA5-66314) and a 180 kDa band corresponding to SRC2 was observed across all tested cell line lysates, along with uncharacterized bands at 110 kDa, 55 kDa and 40 kDa. Whole cell extracts (30 µg lysate) of HEK-293 (Lane 1), HeLa (Lane 2) and A549 (Lane 3) prepared using RIPA lysis buffer were electrophoresed using NuPAGE™ 3-8% Tris-Acetate Protein Gel (Product # EA0378BOX), 10 well. Resolved proteins were equilibrated with 20% ethanol and then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1 µg/mL dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:10,000 dilution) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Pico PLUS Chemiluminescent Substrate (Product # 34580).
