Fluorescent western blot was performed using Rabbit anti-Mouse IgG2b Recombinant Secondary Antibody (Product # SA5-10210). Whole cell extracts of A-431 (Lane 1, 2, 3) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). The blot was probed with Cytokeratin 17 Monoclonal Antibody (Product # MA1-06325) and GAPDH Monoclonal Antibody (Product # MA5-15738). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). Secondary antibodies (Product # SA5-10210, 0.2 µg/mL) and (Product # A32723, 1:5,000) were used for detection of Cytokeratin 17 and GAPDH respectively. Goat anti-Rabbit IgG (H+L) Highly Cross-Adsorbed Secondary Antibody, Alexa Fluor™ 790 (Product # A11369, 1:5,000) was used as a tertiary antibody to detect Rabbit anti-Mouse IgG2b Recombinant Secondary Antibody using the iBright™ FL 1500 (Product # A44115). Rabbit anti-Mouse IgG2b Recombinant Secondary Antibody (Product # SA5-10210) specifically detects the Cytokeratin 17 primary antibody.
