Western Blot was performed using Anti-Perforin Monoclonal Antibody (eBioBOR21), eBioscience™ (Product # 14-9993-82), and a 61 kDa band corresponding to Perforin-1 was observed in the NK-92 cell line but not in the A-431 cell line as reported. An uncharacterized band (*) at ~110 kDa was also observed in the A-431 cell line. Whole-cell extracts (30 µg lysate) of NK-92 (Lane 1), NK-92 treated with PTI (1X for 4h) (Lane 2), A-431 (Lane 3) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The Blot was probed with the primary antibody (2 µg/mL) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076).
