The NFkB/Rel transcription factors are present in the cytosol in an inactive state, complexed with the inhibitory IkB proteins. Most agents that activate NFkB do so through a common pathway based on phosphorylation-induced, proteasome-mediated degradation of IkB. The key regulatory step in this pathway involves activation of a high molecular weight IkB kinase (IKK) complex, whose catalysis is generally carried out by three tightly associated IKK subunits. IKKa and IKKb serve as the catalytic subunits of the kinase. IKKg serves as its regulatory subunit. Activation of IKK depends on phosphorylation; serines 177 and 181 in the activation loop of IKKb (176 and 180 in IKKa) are the specific sites whose phosphorylation causes a conformational change that results in kinase activation
