Pulsed Field Gel Electrophoresis Systems Pulsed-field gel electrophoresis (PFGE) systems allow for the separation of large DNA fragments, reaching upwards into the megabase range. In contrast to standard DNA gel electrophoresis systems, PFGE applies an electric field that periodically changes direction, causing large DNA molecules to move in a zigzag pattern. Depending on the PFGE system, DNA fragments can be resolved at sizes from 100 bp to 10 Mb. When evaluating PFGE systems, consider the electrophoresis angle. In contrast to an angle of 120°, a 90° angle allows DNA movement of four times per cycle instead of the standard two.
