T4 DNA polymerase is an enzyme that catalyzes the synthesis of DNA in the 5’ to 3' direction. Originally derived from the bacteriophage T4, the enzyme is recombinantly expressed in E. coli and purified for use in molecular biology. This polymerase is template-dependent, requires a primer, and contains 3' to 5' exonuclease activity. However, unlike DNA polymerase 1, T4 DNA polymerases lack a 5'to 3' exonuclease function. They can be used for site-directed mutagenesis, making blunt ends for 5' or 3' overhangs, probe labeling using replacement synthesis, and sublconing.
