Description

The STELLARIS DIVE multiphoton (MP) microscope offers multicolor imaging in highly scattering samples. STELLARIS DIVE grants flexibility in the choice of fluorescent probes and therefore allows the study of simultaneous events deep within living specimens or tissue. The system expands the potential of experimental setups by adding label-free contrast like Second or Third Harmonic Generation (SHG/THG) on the fly. STELLARIS DIVE is equipped with 4Tune, a spectrally tunable non-descanned detection with up to 4 spectrally tunable channels. The spectral characteristics of each detection window are seamlessly tunable between 380 and 800 nm. This is achieved using variable dichroics and bandpass filters. The most sensitive detection is accessible with GaAsP Hybrid detectors, compared to PMTs. The 4Tune user interface allows users to optimize the emission setting for multiple transgenic markers by simple drag and drop. Due to its clear and intuitive design, operating it is easy and requires minimal training. The 4Tune detection allows users to specifically define a narrow detection window for 2nd or 3rd harmonics anywhere in the spectrum to match the excitation wavelength of a fluorophore of interest. The detection window size can be adjusted regarding the spectral characteristics for optimal separation of fluorescent markers. Emission lambda scans on the non-descanned detector (NDD) are therefore possible too.
