Intended Uses: This human tissue-type plasminogen activator (tPA) total antigen assay is intended for the quantitative determination of total tissue plasminogen activator in human plasma and other biological fluids. For research use only.
Principle of the Assay: Human tPA will bind to the affinity purified capture antibody coated on the microtiter plate. Free, latent and complexed tPA will bind to the plate. After appropriate washing steps, monoclonal anti-human tPA primary antibody binds to the captured protein. Excess primary antibody is washed away and bound antibody is reacted with the peroxidase conjugated secondary antibody. Following an additional washing step, TMB is used for color development at 450nm. A standard calibration curve is prepared along with the samples to be measured using dilutions of human tPA. Color development is proportional to the concentration of tPA in the samples.
Background: tPA is a serine protease that catalyzes the activation of plasminogen to plasmin [1]. Clinical studies have indicated that high tPA levels may increase the risk for thrombosis [2], whereas decreased levels may cause neuronal plasticity and degeneration [3]
