Description


Principle of the assay: This assay employs a two-site sandwich ELISA to quantitative GPC1 in Human serum, plasma. An antibody specific for GPC1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any GPC1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin - conjugated antibody specific for GPC1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of GPC1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Cell surface heparan sulfate proteoglycans are composed of a membraneassociated protein core substituted with a variable number of heparan sulfate chains. Members of the glypican-related integral membrane proteoglycan family (GRIPS) contain a core protein anchored to the cytoplasmic membrane via a glycosyl phosphatidylinositol linkage. These proteins may play a role in the control of cell division and growth regulation. Moreover, athymic mice lacking Gpc1 exhibited decreased tumor angiogenesis and metastases following intrapancreatic implantation with human pancreatic cancer cells and fewer pulmonary metastases following intravenous injection of mouse melanoma cells
