Description
Principle of the assay: human Galectin-9 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for Galectin-9 has been precoated onto 96-well plates. Standards(E.coli, M1-T355) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Galectin-9 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human Galectin-9 amount of sample captured in plate.
Background: Galectin-9, also called HUAT or LGALS9A is a protein that in humans is encoded by the LGALS9 gene. This gene is mapped to 17q11.2. The galectins are a family of beta-galactoside-binding proteins implicated in modulating cell-cell and cell-matrix interactions. The protein encoded by this gene is an S-type lectin. It is overexpressed in Hodgkin's disease tissue and might participate in the interaction between the H&RS cells with their surrounding cells and might thus play a role in the pathogenesis of this disease and/or its associated immunodeficiency. The protein has N- and C- terminal carbohydrate-binding domains connected by a link peptide
