Description

Background: Macrophage inflammatory protein-1 alpha (MIP-1 alpha), also called CCL3, LD78. The cDNA for MIP-1 alpha predicts a mature peptide of 69 amino acids with a molecular mass of 7,889 daltons. LD78 is a member of a newly identified superfamily of small inducible proteins involved in inflammatory responses, wound healing and tumorigenesis. MIP-1 alpha is a chemokine that has pro-inflammatory and stem cell inhibitory activities in vitro. It constitutes an important second signal for mast cell degranulation in the conjunctiva in vivo and consequently for acute-phase disease.

Principle of the Assay: The Rat CCL3 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Rat CCL3 in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Rat CCL3 coated on a 96-well plate. Standards and samples are pipetted into the wells and CCL3 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Rat CCL3 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CCL3 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm
