Principle of the Assay::The Rat CD325 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Rat CD325 in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Rat CD325 coated on a 96-well plate. Standards and samples are pipetted into the wells and CD325 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Rat CD325 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CD325 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
Background/Introduction: Cadherin-2 (CDH2), also known as neural cadherin(NCAD), is a protein that in humans is encoded by the CDH2 gene. It is a classical cadherin from the cadherin superfamily. This gene is mapped to 18q12.1. Cadherin-2 is expressed in the brain, skeletal and cardiac muscle. Cadherin-2 is commonly found in cancer cells and provides a mechanism for transendothelial migration. It is a calcium dependent cell-cell adhesion glycoprotein comprising five extracellular cadherin repeats, a transmembrane region and a highly conserved cytoplasmic tail. The protein functions during gastrulation and is required for establishment of left-right asymmetry. At certain central nervous system synapses, presynaptic to postsynaptic adhesion is mediated at least in part by this gene product
