Principle of the Assay: Macrophage inflammatory protein-1 alpha (MIP-1 alpha), also called CCL3, LD78. The cDNA for MIP-1 alpha predicts a maturepeptide of 69 amino acids with a molecular mass of 7,889 daltons. LD78 is a member of a newly identified superfamily of small inducible proteins involved in inflammatory responses, wound healing and tumorigenesis.Human CCL3 ELISA Kitemploys a double antibody sandwich method to quantitate Human CCL3 in samples. An antibody specific for Human CCL3 hasbeen pre-coated onto a microplate. Standards and samples are pipetted into the wells and any Human CCL3 present is bound bythe immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for Human CCL3 isadded to the wells. After washing, proprietary Streptavidin-HRP conjugates is added to the wells. Following a wash to remove anyunbound streptavidin-enzyme reagent, adding HRP Substrate (TMB), TMB turns blue under the catalysis of HRP, and turns yellowafter adding stop solution. Measure the OD value with a microplate reader at 450 nm wavelength. The Human CCL3concentration is proportional to the OD450 nm value
