Principle of the Assay: TNF-alpha is synthesized as a 26 kDa, type II transmembrane protein that is 233 amino acids in length. It contains a 30 amino acid(aa) cytoplasmic domain, a 26 aa transmembrane segment, and a 177 aa extracellular region. TNF-alpha is assembled intracellularlyto form a transmembrane, non-covalently-linked homotrimeric protein.Mouse TNF-alpha ELISA Kit employs a doubleantibody sandwich method to quantitate Mouse TNF-alpha in samples. An antibody specific for Mouse TNF-alpha has been pre-coatedonto a microplate. Standards and samples are pipetted into the wells and any Mouse TNF-alpha present is bound by the immobilizedantibody. After removing any unbound substances, a biotin-conjugated antibody specific for Mouse TNF-alpha is added to the wells.After washing, proprietary Streptavidin-HRP conjugates is added to the wells. Following a wash to remove any unboundstreptavidin-enzyme reagent, adding HRP Substrate (TMB), TMB turns blue under the catalysis of HRP, and turns yellow afteradding stop solution.
