Description

Background: The 92 kDa type IV collagenase is also known as 92 kDa gelatinase, type V collagenase, gelatinase B, or matrix metalloproteinase-9 (MMP9). The 72 and 92 kDa type IV collagenases are members of a group of secreted zinc metalloproteases. The matrix metalloproteinases (MMPs) are able to degrade the extracellular matrix and allow angiogenesis and tumor invasion. Gelatinase B, a matrix metalloproteinase that has proteolytic activity against connective tissue proteins, has been suggested to be important in the connective tissue remodeling processes associated with atherogenesis and plaque rupture. MMP-9 is predominantly expressed in neutrophils, macrophages, and mast cells, rather than in oncogene-positive neoplastic cells. The polymorphism of MMP-9 acts as a genetic factor for the development of smoking-induced pulmonary emphysema.

Principle of the Assay: The AAA Biotech' Human MMP9 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human MMP9 in Cell Culture Supernatants, Serum, Plasma, Tissue Homogenates. This assay employs an antibody specific for Human MMP9 coated on a 96-well plate. Standards and samples are pipetted into the wells and MMP9 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human MMP9 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of MMP9 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm
