Description

Background: Plasminogen activator, tissue, also called PLAT or TPA is a serine protease. This gene encodes tissue-type plasminogen activator, a secreted serine protease which converts the proenzyme plasminogen to plasmin, a fibrinolytic enzyme. Tissue-type plasminogen activator is synthesized as a single chain which is cleaved by plasmin to a two chain disulfide linked protein. This gene was mapped to 8p11.21. This enzyme plays a role in cell migration and tissue remodeling. Increased enzymatic activity causes hyperfibrinolysis, which manifests as excessive bleeding; decreased activity leads to hypofibrinolysis which can result in thrombosis or embolism.

Principle of the Assay: The Human tPA ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human tPA in Cell Culture Supernatants, Serum, Plasma, Tissue Homogenates. This assay employs an antibody specific for Human tPA coated on a 96-well plate. Standards and samples are pipetted into the wells and tPA present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human tPA antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of tPA bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm
