Principle of the Assay: Human Xaa-Pro Dipeptidase/Prolidase (PEPD) ELISA Kit employs a two-site sandwich ELISA to quantitate PEPD in samples. An antibody specific for PEPD has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PEPD present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Human PEPD detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of PEPD bound in the initial step. The color development is stopped and the intensity of the color is measured.
Background/Introduction: Xaa-Pro dipeptidase is a cytosolic dipeptidase that hydrolyzes dipeptides with proline or hydroxyproline at the carboxy terminus (but not Pro-Pro). It is important in collagen metabolism because of the high levels of aminoacids.The protein forms a homodimer that hydrolyzes dipeptides or tripeptides with C-terminal proline or hydroxyproline residues. The enzyme serves an important role in the recycling of proline, and may be rate limiting for the production of collagen. Mutations in this gene result in prolidase deficiency, which is characterized by the excretion of large amount of di- and tri-peptides containing proline.
