Background: Leptin is a hormone secreted by adipose tissue. Its main function is to regulate body weight and energy balance by affecting appetite and metabolic rate. Leptin acts through receptors in areas such as the hypothalamus and abdomen.The level of leptin usually increases with an increase in body fat, and is also influenced by many other factors, such as diet, exercise, physiology, and disease. Research has shown that both high and low levels of leptin may be associated with weight and energy balance-related issues, such as obesity and metabolic syndrome. Leptin is also related to many other physiological processes, such as immune function, reproductive function, and bone formation. Understanding the role and regulatory mechanisms of leptin is important for understanding the mechanisms of weight regulation, metabolic diseases, and other related diseases.
Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti Leptin antibody was precoated onto the 96-well plate. The biotin conjugated anti Leptin antibody was used as the detection antibody. The standards and pilot samples were added to the wells subsequently. After incubation, unbound conjugates were removed by wash buffer. Then, biotinylated detection antibody was added to bind with Leptin conjugated on coated antibody. After washing off unbound conjugates, HRP-Streptavidin was added. After a third washing, TMB substrates were added to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that turned yellow after adding a stop solution. Read the O.D. absorbance at 450nm in a microplate reader. The concentration of Leptin in the sample was calculated by drawing a standard curve. The concentration of the target substance is proportional to the OD450 value
