Complement proteins, such as C4a, are present at developing synapses during the active phase of synapse elimination, which is crucial for proper brain wiring. In mice, Complement Component 4a (C4a) is primarily involved in eliminating synapses within the central nervous system (CNS), facilitating normal nervous system development. In some neurodegenerative diseases, where synapse loss occurs during early brain maturation, there is a significant increase in the expression of complement components like C4a. This elevation suggests that the mechanisms of complement-mediated synapse elimination may be reactivated during disease progression, potentially driving the advancement of these conditions
