This Human Glial Cell Line Derived Nerutrophic Factor (GDNF) Rapid ELISA Kit is intended is a fast ELISA kit, this assay takes less than 1.5 hours to detect Human GDNF with <4pg/ml sensitivity. format: 96-well plate with removable strips. Compatible samples: cell culture supernates, cell lysates, serum and plasma (heparin, EDTA). This is a TMB colorimetric sandwich ELISA kit with short assay time and fast experiment set up. GDNF tissue specificity: In the brain, predominantly expressed in the striatum with highest levels in the caudate and lowest in the putamen. Isoform 2 is absent from most tissues except for low levels in intestine and kidney. Highest expression of isoform 3 is found in pancreatic islets. Isoform 5 is expressed at very low levels in putamen, nucleus accumbens, prefrontal cortex, amygdala, hypothalamus and intestine. Isoform 3 is up-regulated in the middle temporal gyrus of Alzheimer disease patients while isoform 2 shows no change. Strip well format. Reagents for up to 96 tests.
This human GDNF ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for GDNF has been precoated onto 96-well plates. Standards (NSO, R109-I211) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for GDNF is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human GDNF amount of sample captured in plate.
