Description

This Human Programmed Cell Death 1 Ligand 2 (CD273) ELISA Kit is intended for Quantitative Detection of human PD-L2 in cell culture supernates, serum and plasma (heparin, EDTA). Strip well format. Reagents for up to 96 tests.
This human PD-L2 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for PD-L2 has been precoated onto 96-well plates. Standards (Expression system for standard: NSO, Immunogen sequence: L20-T220) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for PD-L2 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human PD-L2 amount of sample captured in plate.
The capture antibody is a monoclonal antibody from mouse, the detection antibody is a biotinylated polyclonal antibody from goat. Expression system for standard: Programmed cell death 1 ligand 2 (also known as PD-L2, B7-DC) is a protein that in humans is encoded by the PDCD1LG2 gene. It is mapped to chromosome 9p24.2. PD-L2 is one of two ligands for programmed death-1 (PD-1), a member of the CD28 family of immuno-receptors. Northern blot analysis detect wide expression of PDL2, with highest levels in placenta, heart, pancreas, lung, and liver, and lower levels in spleen, lymph nodes, and thymus. PDL2 negatively regulates T cells and plays an essential role in immune tolerance
