This Human Nephrin ELISA Kit is intended for quantitative detection of human Nephrin in cell culture supernates, serum and plasma (heparin, EDTA, citrate). Strip well format. Reagents for up to 96 tests.
This human Nephrin ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for Nephrin has been precoated onto 96-well plates. Standards (Expression system for standard: NSO, Immunogen sequence: Q23-S1055) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Nephrin is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substmousee TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human Fibronectin amount of sample captured in plate.

Expression system for standard: Nephrin is a protein necessary for the proper functioning of the renal filtration barrier. It is mapped to 19q13.12. This gene encodes a member of the immunoglobulin family of cell adhesion molecules that functions in the glomerular filtration barrier in the kidney. The gene is primarily expressed in renal tissues, and the protein is a type-1 transmembrane protein found at the slit diaphragm of glomerular podocytes. The slit diaphragm is thought to function as an ultrafilter to exclude albumin and other plasma macromolecules in the formation of urine. Mutations in this gene result in Finnish-type congenital nephrosis 1, characterized by severe proteinuria and loss of the slit diaphragm and foot processes
