Description

This Human Carbonic Anhydrase 3 ELISA Kit is intended for quantitative detection of human CA3 in cell culture supernates, serum and plasma (heparin, EDTA, citrate) and urine. Strip well format. Reagents for up to 96 tests.
This human CA3 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for CA3 has been precoated onto 96-well plates. Standards (Expression system for standard: E Coli, Immunogen sequence: A2-K260) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for CA3 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human Fibronectin amount of sample captured in plate.
The capture antibody is a monoclonal antibody from mouse, the detection antibody is a biotinylated detection polyclonal antibody from goat. Expression system for standard: Carbonic anhydrase III (CA3) is an enzyme that in humans is encoded by the CA3 gene. CA3 is a member of a multigene family that encode carbonic anhydrase isozymes. It is mapped to 8q21.2. The gene spans 10.3 kb and contains seven exons and six introns. Using a cDNA clone of the CA3 gene in the study of human-rodent hybrids, the gene was mapped to chromosome 8 which carries a cluster of CA genes. The expression of the CA3 gene is strictly tissue specific and present at high levels in skeletal muscle and much lower levels in cardiac and smooth muscle. A proportion of carriers of Duchenne muscle dystrophy have a higher CA3 level than normal
