This human CPA1 ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for CPA1 has been precoated onto 96-well plates. Standards (Expression system for standard: E Coli, Immunogen sequence: A111-Y419) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for CPA1 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB are used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human CPA1 amount of sample captured in plate.

Expression system for standard: Carboxypeptidase A1 is an enzyme that in humans is encoded by the CPA1 gene. It is mapped to 7q32.2. This gene encodes a member of the carboxypeptidase A family of zinc metalloproteases. This enzyme is produced in the pancreas and preferentially cleaves C-terminal branched-chain and aromatic amino acids from dietary proteins. This gene and several family members are present in a gene cluster on chromosome 7. Mutations in this gene may be linked to chronic pancreatitis, while elevated protein levels may be associated with pancreatic cancer
