This Human Cobalamin Binding (Gastric) Intrinsic Factor ELISA Kit is intended for quantitative detection of human GIF in cell culture supernates, serum and plasma (heparin, EDTA, citrate). Strip well format. Reagents for up to 96 tests.
This human GIF ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for GIF has been precoated onto 96-well plates. Standards (Expression system for standard: NSO, Immunogen sequence: S19-Y417) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for GIF is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB are used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human GIF amount of sample captured in plate.

Expression system for standard: Intrinsic factor (IF), also known as gastric intrinsic factor (GIF), is a glycoprotein produced by the parietal cells of the stomach. It is necessary for the absorption of vitamin B12 later on in the ileum of the small intestine. In humans, the gastric intrinsic factor protein is encoded by the GIF gene. It is mapped to 11q12.1. Haptocorrin (also known as HC, R protein, and transcobalamin I, TCN1) is another glycoprotein secreted by the salivary glands which binds to vitamin B12. Vitamin B12 is acid sensitive and in binding to transcobalamin I it can safely pass through the acidic stomach to the duodenum. In the less acidic environment of the small intestine, pancreatic enzymes digest the glycoprotein carrier and vitamin B12 can then bind to intrinsic factor
