This Human Prostate-Specific Antigen (KLK3) ELISA Kit is intended for quantitative detection of Human Kallikrein 3 in cell culture supernates, cell lysates, serum and plasma (heparin, EDTA). Strip well format. Reagents for up to 96 tests.
This human Kallikrein 3 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for Kallikrein 3 has been precoated onto 96-well plates. Standards (NSO, A18-P261) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Kallikrein 3 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human Kallikrein 3 amount of sample captured in plate.
The capture antibody is a monoclonal antibody from mouse, the detection antibody is a biotinylated polyclonal antibody from goat. Expression system for standard: Prostate-specific antigen (PSA), also known as kallikrein III, seminin, semenogelase, gamma-seminoprotein and P-30 antigen) is a 34 kD glycoprotein manufactured almost exclusively by the prostate gland; PSA is produced for the ejaculate where it liquifies the semen in the seminal coagulum and allows sperm to swim freely.1It is also believed to be instrumental in dissolving the cervical mucous cap, allowing the entry of sperm.2It is a serine protease enzyme, the gene of which is located on the nineteenth chromosome (19q13).3
