Description

Description: This assay employs a two-site sandwich ELISA to quantitate CDH4 in samples. An antibody specific for CDH4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CDH4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CDH4 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CDH4 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: Cadherin-4 is a classical cadherin from the cadherin superfamily. The encoded protein is a calcium-dependent cell-cell adhesion glycoprotein composed of five extracellular cadherin repeats, a transmembrane region and a highly conserved cytoplasmic tail. Based on studies in chicken and mouse, this cadherin is thought to play an important role during brain segmentation and neuronal outgrowth.In addition, a role in kidney and muscle development is indicated. Of particular interest are studies showing stable cis-heterodimers of cadherins 2 and 4 in cotransfected cell lines.
