Description: This assay employs a two-site sandwich ELISA to quantitate CDH19 in samples. An antibody specific for CDH19 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CDH19 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CDH19 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CDH19 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: CDH19 is a type II classical cadherin from the cadherin superfamily and one of three cadherin 7-like genes located in a cluster on chromosome 18. The encoded membrane protein is a calcium dependent cell-cell adhesion glycoprotein comprised of five extracellular cadherin repeats, a transmembrane region and a highly conserved cytoplasmic tail. Type II (atypical) cadherins are defined based on their lack of a HAV cell adhesion recognition sequence specific to type I cadherins.
