Description

Description: This assay employs a two-site sandwich ELISA to quantitate EFNB3 in samples. An antibody specific for EFNB3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any EFNB3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for EFNB3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of EFNB3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: EFNB3, a member of the ephrin gene family, is important in brain development as well as in its maintenance. Moreover, since levels of EFNB3 expression were particularly high in several forebrain subregions compared to other brain subregions, it may play a pivotal role in forebrain function. The EPH and EPH-related receptors comprise the largest subfamily of receptor protein-tyrosine kinases and have been implicated in mediating developmental events, particularly in the nervous system. EPH Receptors typically have a single kinase domain and an extracellular region containing a Cys-rich domain and 2 fibronectin type III repeats. The ephrin ligands and receptors have been named by the Eph Nomenclature Committee (1997)
