Description

Description: This assay employs a two-site sandwich ELISA to quantitate MEX3D in samples. An antibody specific for MEX3D has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MEX3D present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MEX3D is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MEX3D bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: MEX3D is an RNA binding protein that interacts with AU-rich elements of Bcl-2. Upon binding, MEX3D has a negative regulatory action on Bcl-2 expression at the posttranscriptional level.MEX3 proteins contain two N-terminal heterogeneous nuclear ribonucleoprotein K homology motifs and a RING domain at the C-terminus.The predicted 490-amino acid partial protein has 2 N-terminal ribonucleoprotein K homology (KH) domains and a C-terminal RING domain, and it shares similarity with C. elegans Mex3. Northern blot analysis detected a 3.1-kb transcript in all tissues examined, with highest expression in placenta. A 6.5-kb transcript was also present in heart and skeletal muscle. Immunofluorescence analysis of HeLa cells localized endogenous TINO predominantly in the nucleus and perinuclear region
