Description

Description: This assay employs a two-site sandwich ELISA to quantitate HIST1H2AC in samples. An antibody specific for HIST1H2AC has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any HIST1H2AC present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for HIST1H2AC is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of HIST1H2AC bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Two molecules of each of the four core histones (H2A, H2B, H3, and H4) form an octamer, around which approximately 146 bp of DNA is wrapped in repeating units, called nucleosomes. The linker histone, H1, interacts with linker DNA between nucleosomes and functions in the compaction of chromatin into higher order structures.
HIST1H2AC is intronless and encodes a member of the histone H2A family. Transcripts from this gene lack polyA tails but instead contain a palindromic termination element. This gene is found in the large histone gene cluster on chromosome 6
